Gene Taq FP

Thermus aquaticus YT1のDNAポリメラ−ゼ遺伝子をクロ−ニングし、改変したものを大腸菌にて発現させ、分離し、高度に精製した耐熱性DNAポリメラ−ゼである。天然型Taq DNAポリメラ−ゼのN末端を一部欠失させてあるため、内因性の5'→3'エキソヌクレア−ゼ活性を有していない。

	[単位の定義]	1unitsは、activated calf thymus DNAをプライマー/鋳型として74°C、30分間に10nmoleのデオキシヌクレオチドを酸不溶性沈殿物に取り込む酵素活性とする。
	[形状]	20 mmol/l Tris-HCl(pH8.0), 100 mmol/l KCl, 0.1 mmol/l EDTA, 0.5% Tween20, 1 mmol/l DTT, 50% Glycerol
	[純度]	本酵素10 unitsと1µgのλ/Hind III digestとを74°C、1時間反応させてもアガロースゲル電気泳動パターンに変化は認められない。
	[dNTP Mixture]	250 unitsごとに1本(800µl)添付 2.5 mmol/l dATP, 2.5 mmol/l dTTP, 2.5 mmol/l dGTP, 2.5 mmol/l dCTP
	[10×Gene Taq Universal Buffer]	250 unitsごとに1本(1ml)添付 Mg2+濃度 : 15 mmol/l
