mRNA Fingerprinting Kit Ver.1.0SGは、真核生物のmRNA フィンガープリントを作製するためのキットです。アイソトープを用いず、また特別な装置を使用せずに、再現性の高いmRNAフィンガープリント像を得られるようデザインされています。

[mRNA Fingerprinting Kit Ver.1.0SGの原理]

1. RNA抽出用試薬『ISOGEN』等を用いて、DNAの混入していないtotal RNAを準備します。
2. poly(A)⁺ RNAのpoly A 配列にアニールするように設計された3'-Anchor Primerを用いて逆転写反応を行い、cDNAを合成します。
3. 任意の10mer配列からなるArbitrary Primerと3'-Anchor Primer によるPCRを行います。
4. PCR産物を未変性ポリアクリルアミドゲル電気泳動し、SYBR® Green I染色し、紫外線を照射してバンドを検出します。
5. 目的DNAバンドを含むゲルを切り出し、同じArbitrary Primerと3'-Anchor Primerを用いて再増幅し、この後クローニングして解析します。

[使用上の注意]

• 本品では手軽に行える未変性ポリアクリルアミドゲルをSYBR® Green Iにて染色する方法を採用しています。従って従来のアイソトープや蛍光プローブで標識したプライマーを用いた方法よりゲル1レーン当たりのバンド数は少なくなります。
• 本品を用いて検出できるバンドの数及び再現性は、用いるtotal RNAの純度や使用するサーマルサイクラーによっても影響を受けます。常に高純度のtotal RNAを使用し、同一のサーマルサイクラーと、同一のチューブを使用することが必要です。
• ゲル上で検出したフィンガープリント像から、シグナル強度の差が検出されたDNAバンドは、必ずしも真の遺伝子の発現量の差や発現の有無を反映するとは限りません。特に個体間の比較をする場合、多型を十分考慮する必要があります。一般的に、発現量に差があると思われるDNAバンドを切り出し、クローニング後、様々な解析を行い、真に差のある遺伝子であると決定されるわけですが、これらの操作は、非常に時間がかかることをご承知ください。
• PCRをベースとした本法において、発現に差のある遺伝子全てを検出することは困難であると思われます。mRNAの存在比に由来するPCRの再現性、非常に少ないmRNAの検出限界等まだよく解明されていない問題が存在することに注意する必要があります。
• 再現性のある結果を得るためには、正確なピペットハンドリングが必要になる場合もあります。使用する自動ピペット(例えばピペットマン、フィンピペット等)の、保証精度を考慮に入れて操作をすることをお勧めします。
